Recent Advances in the Biological Investigation of Organometallic Platinum-Group Metal (Ir, Ru, Rh, Os, Pd, Pt) Complexes as Antimalarial Agents
In the face of the recent pandemic and emergence of infectious diseases of viral origin, research on parasitic diseases such as malaria continues to remain critical and innovative methods are required to target the rising widespread resistance that renders conventional therapies unusable. The prolific use of auxiliary metallo-fragments has augmented the search for novel drug regimens in an attempt to combat rising resistance. The development of organometallic compounds (those containing metal-carbon bonds) as antimalarial drugs has been exemplified by the clinical development of ferroquine in the nascent field of Bioorganometallic Chemistry. With their inherent physicochemical properties, organometallic complexes can modulate the discipline of chemical biology by proffering different modes of action and targeting various enzymes. With the beneficiation of platinum group metals (PGMs) in mind, this review aims to describe recent studies on the antimalarial activity of PGM-based organometallic complexes. This review does not provide an exhaustive coverage of the literature but focusses on recent advances of bioorganometallic antimalarial drug leads, including a brief mention of recent trends comprising interactions with biomolecules such as heme and intracellular catalysis. This resource can be used in parallel with complementary reviews on metal-based complexes tested against malaria.
Cytotoxicity Evaluation of Unmodified Paddlewheel Dirhodium(II,II)-Acetate/-Formamidinate Complexes and Their Axially Modified Low-Valent Metallodendrimers
Two diphenyl formamidine ligands, four dirhodium(II,II) complexes, and three axially modified low-valent dirhodium(II,II) metallodendrimers were synthesized and evaluated as anticancer agents against the A2780, A2780 , and OVCAR-3 human ovarian cancer cell lines. The dirhodium(II,II) complexes show moderate cytotoxic activity in the tested tumor cell lines, with acetate and methyl-substituted formamidinate compounds displaying increased cytotoxicity that is relative to cisplatin in the A2780 cisplatin resistant cell line. Additionally, methyl- and fluoro-substituted formamidinate complexes showed comparable and increased cytotoxic activity in the OVCAR-3 cell line when compared to cisplatin. The low-valent metallodendrimers show some activity, but a general decrease in cytotoxicity was observed when compared to the precursor complexes in all but one case, which is where the more active acetate-derived metallodendrimer showed a lower IC value in the OVCAR-3 cell line in comparison with the dirhodium(II,II) tetraacetate.
Antiplasmodial Activity and In Vivo Bio-Distribution of Chloroquine Molecules Released with a 4-(4-Ethynylphenyl)-Triazole Moiety from Organometallo-Cobalamins
We have explored the possibility of using organometallic derivatives of cobalamin as a scaffold for the delivery of the same antimalarial drug to both erythro- and hepatocytes. This hybrid molecule approach, intended as a possible tool for the development of multi-stage antimalarial agents, pivots on the preparation of azide-functionalized drugs which, after coupling to the vitamin, are released with a 4-(4-ethynylphenyl)-triazole functionality. Three chloroquine and one imidazolopiperazine derivative (based on the KAF156 structure) were selected as model drugs. One hybrid chloroquine conjugate was extensively studied via fluorescent labelling for in vitro and in vivo bio-distribution studies and gave proof-of-concept for the design. It showed no toxicity in vivo (zebrafish model) as well as no hepatotoxicity, no cardiotoxicity or developmental toxicity of the embryos. All 4-(4-ethynylphenyl)-triazole derivatives of chloroquine were equally active against chloroquine-resistant (CQR) and chloroquine-sensitive (CQS) strains.
Bicontinuous structured liquids with sub-micrometre domains using nanoparticle surfactants
Bicontinuous jammed emulsions (or bijels) are tortuous, interconnected structures of two immiscible liquids, kinetically trapped by colloidal particles that are irreversibly bound to the oil-water interface. A wealth of applications has been proposed for bijels in catalysis, energy storage and molecular encapsulation, but large domain sizes (on the order of 5 µm or larger) and difficulty in fabrication pose major barriers to their use. Here, we show that bijels with sub-micrometre domains can be formed via homogenization, rather than spinodal decomposition. We achieve this by using nanoparticle surfactants: polymers and nanoparticles of complementary functionality (for example, ion-pairing) that bind to one another at the oil-water interface. This allows the stabilization of the bijel far from the demixing point of the liquids, with interfacial tensions on the order of 20 mN m . Furthermore, our strategy is extremely versatile, as solvent, nanoparticle and ligand can all be varied.

Bilayer self-assembly on a hydrophilic, deterministically nanopatterned surface
We present measurements of the in situ, microscopic architecture of a self- assembled bilayer at the interface between a regularly nanopatterned surface and an aqueous sub-phase using neutron reflectometry. The substrate is patterned with a rectangular array of nanoscale holes. Because of the high quality of the pattern, using neutron reflectometry, we are able to map the surface-normal density distribution of the patterned silicon, the penetration of water into the pattern, and the distribution of a deposited film inside and outside of the etched holes. In this stud; 1,2-dilauroyl-sn-glycero-3-phosphocholine (DLPC) single bilayers were deposited on the hydrophilic patterned surface. For bilayers deposited either by vesicle fusion (VF) or by the Langmuir-Schaefer (L-S) technique, the most consistent model found to fit the data shows that the lipids form bilayer coatings on top of the substrate as well as the bottoms of the holes in an essentially conformal fashion. However, while there is a single bilayer on the unetched silicon surface, the lipids coating the bottoms of the holes form a complex bimodal structure consistent with a rough surface produced by the etching process. This study provides insight into film transfer both outside and inside regular nanopatterned features.
Experimental evaluation of a novel modification of anionic guar gum with maleic anhydride for fracturing fluid
The content of insoluble substance in guar gum solution is high and its temperature resistance is poor. A novel anionic guar gum (GG) derivative was developed and grafted by modification with maleic anhydride. Compared with guar gum, the insoluble content of MAG solution decreased from 13 to 5%, the decomposition temperature of MAG increased from 285 to 308, and the viscoelasticity of MAG gel has been enhanced, especially elasticity. The rheological tests show that the corresponding fracturing fluid belongs to a non-Newtonian pseudoplastic one and the viscosity could maintain 50 mPa∙s for 2 h under conditions of 150 °C and 170 s −1 , which can be applied in a high-temperature well. It is noteworthy that the synthetic protocol of maleate esters guar gum (MAG) is easily available, low cost, and practical. It is conducive to the industrial application of MAG. The work showed a great significance to the modification of GG with acid anhydride for fracturing fluid.
Examining the Role of Implicit Emotional Judgments in Social Anxiety and Experiential Avoidance
This study sought to examine the relationship of implicit emotional judgments with experiential avoidance (EA) and social anxiety. A sample of 61 college students completed the Emotional Judgment – Implicit Relational Assessment Procedure (EJ-IRAP) as well as a public speaking challenge. Implicit judgments were related to greater self-reported EA, anxiety sensitivity, emotional judgments and social anxiety as well as lower performance ratings and willingness in the public speaking challenge. Effects differed by trial type with “Anxiety is bad” biases related to greater EA/anxiety, while “calm is bad” biases related to lower EA/anxiety (“Good” biases were generally unrelated to outcomes). Implicit emotional judgments moderated the relationship of heart rate during the speech with speech time and willingness, such that increases in heart rate were only related to lower speech time and willingness among those high in implicit judgments. Implicit judgments predicted social anxiety above and beyond self-report EA measures. Implicit emotional judgments appear to have a functional role in EA and anxiety that warrants further research.
Assessment of medical student burnout: toward an implicit measure to address current issues
The feasibility of implicitly assessing medical student burnout was explored, using the Implicit Relational Assessment Procedure (IRAP), to measure longitudinal student burnout over the first two years of medical school and directly comparing it with an existing explicit measure of burnout (Maslach Burnout Inventory; MBI). Three successive cohorts of medical students completed both implicit and explicit measures of burnout at several time points during their first two years of medical school. Both assessments were conducted via the internet within a one-week period during the first week of medical school, the end of the first year of medical school, and the end of the second year, though not all cohorts were able to complete the assessments at all time points. Mixed linear models were used to compare the two measures directly, as well as to evaluate changes over time in each measure separately. Minimal correspondence was observed between the implicit and explicit measures of burnout on a within-subject basis. However, when analyzed separately, all subscales of both measures detected significant change over time in the direction of greater levels of burnout, particularly during the first year of medical school. These results provide preliminary evidence the IRAP is able to assess implicit attitudes related to burnout among medical students, though additional research is needed. The IRAP detected consistent improvements in positive implicit attitudes toward medical training during students’ second year of medical school, which was not detected by the MBI. Possible implications of these findings are discussed.
